In aqueous solution the molecule has a bunch of freely-available hydrogen bond donors/acceptors because it’s like. A cyclic peptide chain. Which makes it highly water-soluble, but it’s not going to be able to cross a lipid membrane. But the molecule can change its conformation such that the hydrogen bond donors/acceptors are masked by interacting with each other and the molecule can permeate lipid membranes
